Target Information
In mice, POMC serves various functions, including the regulation of energy balance, appetite, immune response, reproductive system development, and neuroendocrine function. The POMC gene is expressed in the arcuate nucleus of the mouse hypothalamus, where it influences appetite through its derived peptides like α-melanocyte stimulating hormone (α-MSH) and γ-melanocyte stimulating hormone (γ-MSH). Research indicates that POMC neurons are direct targets of leptin, which reduces appetite by activating these neurons. Additionally, the activity of POMC neurons is modulated by the phosphatidylinositol 3-kinase (PI3K) signaling pathway; inhibiting this pathway diminishes the appetite-suppressing effects of leptin and insulin. The electrical activity of POMC neurons is also influenced by neurotransmitters such as orexin/hypocretin, which alter neuronal excitability to affect appetite.
