Target Information
Presenilin-1 serves as a catalytic subunit of the gamma-secretase complex, an endoprotease involved in the intramembrane cleavage of integral membrane proteins, including Notch receptors and amyloid-beta precursor protein (APP). Its protease activity depends on the presence of the other gamma-secretase complex members. In mice, the function of presenilin-1 (PS1) is primarily associated with Alzheimer's disease (AD)-related pathological processes, such as amyloid deposition, neuronal differentiation, and the regulation of calcium signaling pathways. Mutations in the PS1 gene are major contributors to early-onset familial Alzheimer's disease. Research indicates that PS1 mutations elevate the production of amyloid β protein (Aβ), a hallmark of AD pathology. For instance, in the APP/PS1 transgenic mouse model, PS1 mutations significantly increase Aβ deposition, leading to neuronal dysfunction and cognitive deficits.
