Target Information
Pappalysin-1 is a metalloproteinase that specifically cleaves IGFBP-4 and IGFBP-5, releasing bound IGF. The presence of IGF greatly enhances the cleavage of IGFBP-4, while it slightly inhibits the cleavage of IGFBP-5. Isoform 2 cleaves IGFBP-4 much more slowly than PAPP-A, but its ability to cleave IGFBP-5 remains unaffected. Mice with a knockout of the PAPPA gene exhibit a significantly extended lifespan and healthspan, living on average 30%-40% longer than wild-type mice, without any abnormalities in dietary intake or endocrine function. Additionally, PAPPA-deficient mice experience lower tumor incidence and fewer age-related lesions as they age.
