Target Information
Podocalyxin plays a crucial role in regulating adhesion, cell morphology, and cancer progression. As an anti-adhesive molecule, it helps maintain an open filtration pathway between adjacent podocyte foot processes through charge repulsion. Conversely, it also acts as a pro-adhesive molecule, promoting cell adherence to immobilized ligands, and increasing migration rates and cell-cell contacts in an integrin-dependent manner. In mice, Podocalyxin is essential for maintaining the integrity of various barriers, including the glomerular filtration barrier, vascular barrier, and lung barrier. It is also involved in regulating hematopoietic stem cell function and contributing to cancer development. In the pulmonary vasculature, Podocalyxin ensures the integrity of this system by regulating interactions between the extracellular matrix and the basement membrane. Mice deficient in Podocalyxin exhibit increased pulmonary vascular permeability, which results in pulmonary fluid accumulation and impaired lung function.
