Target Information

Tissue-type plasminogen activator (tPA) facilitates the conversion of inactive plasminogen to plasmin by hydrolyzing a specific Arg-Val bond in plasminogen. Through the regulation of plasmin-mediated proteolysis, tPA is crucial in tissue remodeling and degradation, cell migration, and numerous other physiological and pathological processes. During oocyte activation, tPA is involved in the cortical granule reaction in the zona reaction, contributing to the prevention of polyspermy. In mice, tPA has diverse and complex roles across several areas, including reproduction, coagulation, immunity, the nervous system, and tumors. Specifically, tPA is vital in the reproductive process; studies show that mice lacking the tPA gene exhibit reduced efficiency in dissolving the follicular wall during estrus, leading to decreased ovulation efficiency. However, this defect can be partially offset by the functional redundancy of other plasminogen activators, such as urokinase-type plasminogen activator (uPA).
