Target Information
Plasmin, a serine protease activated from plasminogen, is crucial in thrombolysis, breaking down clots by degrading fibrin. Antiplasmin (α2-antiplasmin) serves as a primary inhibitor of plasmin, rapidly binding to plasmin in a stable 1:1 molar ratio to inhibit its activity. In mice, α2-antiplasmin is the primary plasma inhibitor of plasmin, and knockout of its gene (α2-AP−/−) results in increased plasmin activity without causing significant bleeding tendencies. The formation of the Plasmin-antiplasmin complex is an important indicator of plasmin activity and is related to thrombus stability. In α2-AP−/− mice, the enhanced plasmin activity leads to easier thrombus dissolution, reducing the severity of pulmonary embolism.
