Target Information
Histone lysine demethylase PHF8 is an enzyme that specifically targets the di- and monomethyl states of histones, playing a crucial role in cell cycle progression, rDNA transcription, and brain development. It demethylates the mono- and dimethylated histone H3 'Lys-9' residue (H3K9Me1 and H3K9Me2), the dimethylated H3 'Lys-27' (H3K27Me2), and the monomethylated histone H4 'Lys-20' residue (H4K20Me1). In mice, PHF8 is highly expressed in various brain regions, including the prefrontal cortex, striatum, and hippocampus, which are important for emotional and cognitive functions. Mice lacking PHF8 exhibit significant resistance to stress-induced anxiety and depression-like behaviors, suggesting that PHF8 may influence these conditions by regulating receptors in the serotonin signaling pathway, such as Htr1a and Htr2a.
