Target Information

Peroxisome proliferator-activated receptor gamma (PPAR-γ) is a nuclear receptor that binds to peroxisome proliferators, including hypolipidemic drugs and fatty acids. Upon activation by a ligand, PPAR-γ binds to specific DNA sequences known as PPAR response elements (PPRE), modulating the transcription of target genes such as acyl-CoA oxidase. This process regulates the peroxisomal beta-oxidation pathway of fatty acids. In mice, PPAR-γ has wide-ranging and complex functions, impacting metabolism, immunity, inflammation, cardiovascular protection, neuroprotection, bone metabolism, and tumor suppression. Within the immune system, PPAR-γ plays a crucial role by inhibiting proinflammatory cytokine expression and promoting anti-inflammatory responses. In an experimental colitis model, PPAR-γ-deficient mice exhibit more severe inflammatory responses. Following brain injury, PPAR-γ helps reduce inflammation by promoting macrophage polarization to the M2 type. In mice infected with the influenza virus, downregulation of PPAR-γ increases susceptibility to infection, whereas its activation reduces viral replication and the inflammatory response.
