Target Information

Peptidyl-prolyl cis-trans isomerase NIMA-interacting 1 is a PPIase that specifically binds to and isomerizes phosphorylated Ser/Thr-Pro (pSer/Thr-Pro) motifs. By inducing conformational changes in a subset of phosphorylated proteins, it serves as a molecular switch for various cellular processes, displaying a preference for an acidic residue N-terminal to the isomerized proline bond. Pin1−/− mice exhibit notable cell proliferation abnormalities, including weight loss, testicular atrophy, retinal degeneration, and mammary tissue proliferation disorders during pregnancy. These phenotypes closely resemble those seen in mice with cyclin D1 functional deficiency. Research has shown that Pin1 directly interacts with and stabilizes cyclin D1, which is phosphorylated at Thr-286-Pro, enhancing its stability and transcriptional activity within the cell nucleus, thereby regulating cyclin D1 function at both transcriptional and post-translational levels.
