In mice, PADI3 is primarily involved in hair formation and maintaining the epidermal barrier's integrity. Mutations in the PADI3 gene are significantly associated with various diseases. This enzyme is vital for the development of hair follicles, with its expression observed in the medulla and three papillary layers of mouse hair follicles, highlighting its pivotal role in hair formation. Unlike other members of the PADI family, such as PADI1 and PADI2, PADI3 exhibits distinct tissue distribution and functional characteristics.
