Target Information

The carboxylated form of osteocalcin is a primary organic component of the bone matrix, making up 1-2% of total bone protein. It functions as a negative regulator of bone formation, necessary to limit bone growth without affecting bone resorption or mineralization. In contrast, the uncarboxylated form acts as a hormone released by osteoblasts, regulating various cellular processes such as energy metabolism, male fertility, and brain development. Osteocalcin-deficient mice tend to show signs of obesity, increased blood glucose and lipid levels, decreased insulin levels, and insensitivity to glucose and insulin. When the carboxylation function of osteocalcin is inhibited, these mice's metabolic phenotype returns to normal, emphasizing that osteocalcin functions through its uncarboxylated form.
