The kappa-type opioid receptor is a G-protein coupled receptor that primarily binds to endogenous alpha-neoendorphins and dynorphins, though it has a lower affinity for beta-endorphins. It also acts as a receptor for certain synthetic opioids and the psychoactive compound salvinorin A. OPRK1 is significantly associated with alcohol dependence and nicotine addiction, with research indicating that polymorphisms in the OPRK1 gene can influence alcohol preference in mice. This receptor is crucial for pain perception and regulation. Studies using the chronic constriction injury (CCI) model have demonstrated notable changes in OPRK1 expression: a significant increase in the prefrontal cortex (PFC) and a marked decrease in the amygdala (ACC).
