Nucleotide-binding oligomerization domain-containing protein 1 (NOD1) is a pattern recognition receptor (PRR) that identifies bacterial peptidoglycan fragments and other danger signals, playing a role in both innate and adaptive immune responses. In mice, deletion of NOD1 specifically in intestinal epithelial cells results in anxiety-like behaviors and cognitive dysfunction under stress, linked to decreased expression of the glucocorticoid receptor (GR) and melanocortin 1 receptor (MC4R) in the hippocampus. Additionally, the absence of NOD1 is correlated with increased intestinal permeability and changes in the serotonin signaling pathway. These observations underscore the significance of NOD1 in regulating gut-brain axis communication.
