Target Information
Nuclear respiratory factor 1 (Nrf1) is a transcription factor that activates the expression of the EIF2S1 (EIF2-alpha) gene, linking the transcriptional regulation of key metabolic genes to cellular growth and development. It is involved in controlling nuclear genes essential for respiration, heme biosynthesis, and the transcription and replication of mitochondrial DNA. In mice, Nrf1 serves several critical functions, including the regulation of embryonic hematopoiesis, antioxidant response, metabolic balance, neuroprotection, cardiac regeneration, and germ cell development. Nrf1 knockout mice exhibit impaired embryonic hematopoiesis, resulting in anemia and embryonic lethality. Research has demonstrated that Nrf1 is pivotal in fetal liver hematopoiesis, with its absence leading to erythropoiesis defects, subsequently causing anemia and embryonic death. Additionally, while Nrf1 is widely expressed during embryonic development, its role in specific tissues, such as hematopoietic cells, is likely indirect through interactions with interstitial cells.
