Neuronal pentraxin-2 (Nptx2) likely plays a role in altering cellular properties that support long-term plasticity. It adheres to agar matrix in a manner dependent on calcium. Nptx2 reduces the activation of the complement cascade by binding to complement C1q, thus diminishing synaptic loss and neuronal toxicity mediated by microglia. In mouse models, a deficiency in Nptx2 results in elevated complement activity, increased C1q-dependent microglial phagocytosis, and the loss of excitatory synapses, phenomena that are closely linked to the development of neurodegenerative diseases like Alzheimer's disease and frontotemporal dementia.
