Target Information

Neurogenin-3 functions as a transcriptional regulator and, alongside NKX2-2, initiates the transcriptional activation of NEUROD1. It plays a role in neurogenesis and is essential for specifying a common precursor to the four pancreatic endocrine cell types. In mice, Neurogenin-3 (Ngn3) is primarily involved in the development of pancreatic endocrine cells and the formation and regulation of hypothalamic neuroendocrine cells. A deficiency in Ngn3 leads mice to rapidly develop obesity symptoms due to the loss of Pomc (melanocyte-stimulating hormone) expression in the hypothalamus. Specifically, Ngn3 deficiency results in incomplete differentiation of neurons expressing Pomc and Cart (an orexigenic hormone) in the arcuate nucleus, leading to overeating and decreased energy expenditure.
