Target Information

Neurofilaments typically comprise three intermediate filament proteins: NEFL, NEFM, and NEFH, which play a crucial role in maintaining neuronal caliber. They may also collaborate with neuronal intermediate filament proteins PRPH and INA to create neuronal filamentous networks. In mice, the neurofilament light polypeptide (NfL) is essential for the proper formation and maintenance of these neurofilament networks. In a healthy state, NfL helps maintain nerve fibers' structural integrity and supports nerve signal transmission by forming intermediate filaments alongside other neurofilament proteins. However, mutations in the NfL gene, such as the N98S mutation, can disrupt neurofilament networks and result in Charcot-Marie-Tooth disease (CMT) type 2E.
