Target Information

Neprilysin plays a crucial role in breaking down opioid peptides like Met- and Leu-enkephalins by cleaving a Gly-Phe bond. It also catalyzes the cleavage of bradykinin, substance P, and neurotensin peptides. Neprilysin is a key enzyme in degrading Aβ-amyloid protein (Aβ), and its reduced activity is closely linked to the pathology of Alzheimer's disease (AD). Research indicates that decreased neprilysin activity results in elevated Aβ levels in the brain, thus facilitating the onset of AD. For instance, in a mouse model with the neprilysin gene knocked out, there was a notable increase in brain Aβ levels, accompanied by neuronal damage and behavioral deficits.
