Target Information
Nephronectin serves as a functional ligand for integrin alpha-8/beta-1 during kidney development, working to regulate the expression of GDNF in conjunction with integrin alpha-8/beta-1, which is crucial for kidney formation. It may also be involved in the development and functioning of various tissues by regulating cell adhesion, spreading, and survival through interactions with several integrins. In mice, Npnt plays multiple roles, including in kidney development, bone metabolism, cancer progression, and heart development. Kidney development in mice starts on embryonic day 11, when ureteral buds emerge from the protrusions of the Wolffian duct and invade the mesodermal mesenchyme (MM). Npnt facilitates the invasion of ureteral buds and kidney organ formation by binding to integrin α8β1, a key step in embryonic kidney development. Moreover, Npnt maintains the glomerular filtration function by regulating the composition and structure of the glomerular basement membrane (GBM). Studies have shown that the absence of Npnt can lead to abnormal distribution of tubular epithelial cells and alterations in tubular structure, thereby impacting kidney function.
