Target Information

Arylamine N-acetyltransferase 1 (NAT-1) plays a critical role in the detoxification of a wide range of hydrazine and arylamine compounds, with isoniazid, 2-aminofluorene, and anisidine serving as its preferred substrates. Beyond its involvement in drug metabolism and the detoxification of exogenous chemicals in mice, NAT-1 is also integral to various biological processes, including insulin resistance, cancer progression, mitochondrial function, and the antioxidant response. This enzyme is ubiquitously expressed across multiple tissues in mice, such as the liver, muscle, and adipose tissue, where it facilitates the metabolism of numerous drugs and exogenous substances. Notably, NAT1 knockout (Nat1 KO) mice develop systemic insulin resistance, which is linked to a marked accumulation of lipids in the liver and muscle. This condition is characterized by elevated levels of triglycerides (TAG) and diacylglycerol (DAG), as well as compromised mitochondrial function, evidenced by reduced mitochondrial respiration and diminished oxidative phosphorylation capacity.
