Target Information
Myotubularin-related protein 9 (MTMR9) functions as an adapter for myotubularin-related phosphatases, enhancing the catalytic activity of MTMR6, particularly toward phosphatidylinositol 3,5-bisphosphate, and increasing MTMR6's binding affinity for phosphorylated phosphatidylinositols. It also positively regulates the catalytic activity of MTMR7. In mice, MTMR9 has diverse roles, including its interaction with MTMR6, regulation of autophagy and apoptosis, immune cell differentiation, and involvement in metabolism and disease processes. Specifically, MTMR9 influences the balance of Th1, Th2, and Th17 cells by modulating the differentiation of CD4+ T cells. Knockout or inhibition of MTMR9 has been shown to promote Th1 cell differentiation while enhancing the activity of key signaling pathways, such as protein kinase B (AKT) and mitogen-activated protein kinase (MAPK), in Th1 cells. These findings underscore the multifaceted role of MTMR9 in immune regulation and cellular signaling.
