Mothers against decapentaplegic homolog 7 (Smad7) acts as an antagonist to TGF-beta type 1 receptor superfamily signaling. It inhibits TGF-beta and activin signaling by associating with their receptors, thereby blocking SMAD2 access. Smad7 is crucial for cardiac development, as its absence can result in defects such as ventricular septal defects, ventricular noncompaction, and outflow tract malformations. In adults, Smad7 mutant mice experience cardiac dysfunction and severe arrhythmias. Additionally, Smad7 is vital for skeletal muscle development; its deficiency leads to reduced muscle mass, altered myofiber types, and diminished regenerative capacity. Moreover, the loss of Smad7 is linked to enhanced myogenin (GDF-8) signaling.
