Target Information
Mothers against decapentaplegic homolog 2 (SMAD2) is a receptor-regulated SMAD (R-SMAD) that functions as an intracellular signal transducer and transcriptional modulator. It is activated by transforming growth factor-beta (TGF-beta) and activin type 1 receptor kinases. SMAD2 binds to the TRE element in the promoter regions of various TGF-beta-regulated genes, and upon forming the SMAD2/SMAD4 complex, it activates transcription. In hepatocytes, SMAD2 primarily serves as a negative regulator, inhibiting hepatocyte proliferation and differentiation and playing a critical role in liver regeneration. In the central nervous system, SMAD2 is vital for neural development and function. Mice lacking SMAD2 exhibit abnormal cerebellar development, including disordered cerebellar cortical lamination, delayed granule cell maturation, and reduced dendritic spines. These abnormalities result in motor coordination disorders and early mortality in mice.
