Mitochondrial Open Reading Frame of The 12S rRNA-c (MOTS-c) is a peptide encoded by mitochondrial DNA that exhibits multifaceted metabolic regulatory effects in mice. This 16-amino-acid peptide promotes metabolic homeostasis, enhances insulin sensitivity, and regulates energy metabolism primarily by activating the AMPK signaling pathway. By activating AMPK (adenosine monophosphate-activated protein kinase), MOTS-c improves insulin sensitivity. Research has demonstrated that MOTS-c can mitigate insulin resistance and obesity caused by a high-fat diet in mice. Moreover, its expression in skeletal muscle is linked to muscle mass and declines with age, emphasizing its crucial role in maintaining muscle health and metabolic balance.
