Microtubule-associated protein 1 light chain 3 beta (MAP1LC3B) is a ubiquitin-like modifier that plays a crucial role in the formation of autophagosomal vacuoles, or autophagosomes. It is involved in mitophagy, helping to regulate mitochondrial quantity and quality by maintaining mitochondria at a basal level to meet cellular energy needs and prevent excessive ROS production. MAP1LC3B is essential for autophagosome membrane formation and interacts with autophagy-related proteins such as Atg5 and Atg7 to facilitate autophagosome formation. In mice, MAP1LC3B expression and functionality are vital for a normal autophagic process. For instance, in renal tubular epithelial cells, the expression of MAP1LC3B is significantly upregulated in AQP11-deficient mice, which is closely associated with renal cyst formation and highlights the role of autophagy in kidney disease. In mouse models, the absence or dysfunction of MAP1LC3B results in abnormal immune responses. For example, Map1lc3b-/- mice exhibit elevated levels of IL-1β and IL-18 during LPS-induced inflammatory responses, linked to defects in autophagic function.
