Target Information

E3 ubiquitin-protein ligase Mdm2 functions as an E3 ubiquitin-protein ligase that facilitates the ubiquitination of p53/TP53, marking it for degradation by the proteasome. By binding to the transcriptional activation domain of p53/TP53 and p73/TP73, Mdm2 inhibits cell cycle arrest and apoptosis mediated by these proteins. Acting as a key negative regulator of p53, Mdm2 forms a complex with p53 to promote its ubiquitination and subsequent degradation. This regulatory mechanism is vital in mouse development and carcinogenesis. Mdm2 knockout mice often face embryonic lethality due to unchecked p53 activity, resulting in cell cycle arrest and apoptosis.
