Target Information
Stromelysin-1, also known as Matrix metalloproteinase-3 (MMP3), is a metalloproteinase with broad substrate specificity, capable of degrading fibronectin, laminin, gelatins of types I, III, IV, and V, collagens III, IV, X, and IX, and cartilage proteoglycans. It also activates various molecules, including growth factors, plasminogen, and other matrix metalloproteinases like MMP9. Released into the extracellular matrix (ECM) as an inactive pro-enzyme, it is activated through the plasmin cascade signaling pathway, and it also acts intracellularly. In mice, MMP3 has extensive and complex roles across various systems, including the lungs, cardiovascular system, bones, metabolism, nerves, and immune system. In acute respiratory distress syndrome (ARDS), the absence of MMP3 can reduce the inflammatory response and help maintain alveolar surfactant function, thus protecting the lungs from damage. However, the loss of MMP3 may also impair lung barrier function. In a viral meningitis model, for instance, increased levels of MMP3 have been linked to disruption of the blood-brain barrier.
