Target Information
MDGA1, a protein with a MAM domain that anchors glycosylphosphatidylinositol, is essential for the radial migration of cortical neurons in the neocortex's superficial layers. It is involved in forming or maintaining inhibitory synapses and may achieve this by inhibiting NLGN2 activity. MDGA1 is predominantly expressed in outer layer neurons within the mouse cortex, particularly in layer II/III cortical neurons that migrate to the surface and settle in the superficial cortex during development. Research indicates that MDGA1 interacts with axon-rich regions via its MAM domain, thus regulating neuron migration and positioning. By interacting with NLGN2, MDGA1 inhibits inhibitory synapse formation, reducing synaptic inhibition in the hippocampus. This inhibition primarily occurs by preventing NLGN2 from binding to presynaptic neurotransmitter receptors, impacting the balance between excitatory and inhibitory activities within the neural network.
