Low density lipoprotein receptor adapter protein 1 serves as an adapter protein, specifically a clathrin-associated sorting protein (CLASP), essential for the efficient endocytosis of the LDL receptor (LDLR) in polarized cells like hepatocytes and lymphocytes, but not in non-polarized cells such as fibroblasts. It may be necessary for LDL binding and internalization, but not for receptor clustering in coated pits. In mice, LDLRAP1 plays vital biological roles, and its absence can lead to various health issues, including lipid metabolism disorders, atherosclerosis, insulin resistance, and metabolic syndrome. Mice lacking LDLRAP1 demonstrate significant atherosclerotic plaque formation when on a high-fat diet, linked to impaired LDLR function.
