Target Information
Loricrin is a prominent protein in the keratinocyte cell envelope. As a major component of the stratum corneum in the epidermis, it plays a vital role in sustaining the skin's barrier function. In mice, the absence or mutation of loricrin leads to alterations in skin phenotypes, although these are typically less severe than in humans. In mice without loricrin, the formation of the skin barrier is delayed during embryonic development, and they are born with congenital erythroderma, characterized by transparent, shiny skin. However, these phenotypes resolve within 4-5 days after birth. Despite the lack of loricrin, the skin barrier function remains intact, suggesting the presence of compensatory mechanisms, such as the upregulation of other keratinocyte envelope components like SPRRP2D, SPRRP2H, and repetin.
