Target Information

The lipolysis-stimulated lipoprotein receptor (LSR) is believed to play a crucial role in the clearance of triglyceride-rich lipoproteins from the blood. It binds to chylomicrons, LDL, and VLDL in the presence of free fatty acids, facilitating their uptake into cells. Additionally, LSR helps maintain epithelial barrier function by recruiting MARVELD2/tricellulin to tricellular tight junctions. LSR is essential for hepatic lipid metabolism and systemic lipid homeostasis, and its dysfunction is closely linked to various metabolic diseases. Under normal conditions, LSR degrades triglyceride-rich lipoproteins through endocytosis, helping to maintain stable levels of cholesterol and triglycerides in the plasma. However, in LSR knockout mice, impaired LSR function leads to a significant increase in plasma triglyceride and cholesterol levels, along with a reduced clearance rate of lipoproteins. This dysfunction is associated with an elevated risk of atherosclerosis and cardiovascular disease, highlighting the critical role of LSR in lipid metabolism and cardiovascular health.
