Target Information
Phosphatidate phosphatase LPIN1 is a magnesium-dependent enzyme that catalyzes the conversion of phosphatidic acid to diacylglycerol, a critical step in the biosynthesis of triglycerides, phosphatidylcholine, and phosphatidylethanolamine. By regulating this process, LPIN1 plays a central role in controlling fatty acid metabolism at multiple levels. In mice, LPIN1 is involved in a wide range of physiological functions, including lipid metabolism, neuroprotection, adipose tissue development, and the function of cardiac and skeletal muscle. Studies on Lpin1fl/fl mice have revealed peripheral neuropathy characterized by slowed nerve conduction velocity, axonal hyperplasia, and reduced myelin thickness, which are linked to impaired differentiation and proliferation of myelin-forming cells. Additionally, cardiac-specific Lpin1 knockout mice do not exhibit significant cardiac dysfunction under normal conditions, but their underlying metabolic abnormalities may indirectly impact cardiac function over time.
