Phosphatidylcholine-sterol acyltransferase, also known as lecithin-cholesterol acyltransferase (LCAT), is a crucial enzyme in the extracellular metabolism of plasma lipoproteins. Primarily synthesized in the liver and secreted into the bloodstream, LCAT catalyzes the conversion of cholesterol and phosphatidylcholines (lecithins) into cholesteryl esters and lysophosphatidylcholines on the surface of high-density lipoproteins (HDLs) and low-density lipoproteins (LDLs). In mice, LCAT plays a central role in cholesterol esterification within HDL and the regulation of lipid metabolism. By converting free cholesterol into cholesteryl esters, LCAT promotes the maturation and functionality of HDL particles. Mice deficient in LCAT exhibit reduced HDL cholesteryl ester content, elevated levels of non-esterified cholesterol, and an abnormal increase in very low-density lipoprotein (VLDL) particles, underscoring its essential role in maintaining lipid homeostasis.
