Target Information
Lamins are intermediate filament proteins that form a filamentous network, serving as key structural components of the nuclear lamina-a fibrous layer located on the nucleoplasmic side of the inner nuclear membrane. Lamin A and C, in particular, contribute to matrix stiffness by influencing the mechanical properties of the nucleus. In mice, Lamin-A/C has diverse and intricate roles, impacting nuclear architecture, bone metabolism, muscle function, immune responses, inflammation regulation, and embryonic development. Notably, Lamin-A/C is critical for bone metabolism. Mice lacking Lamin-A/C exhibit reduced bone formation, increased bone resorption, and lower bone density, closely resembling the characteristics of age-related osteoporosis. These effects are linked to impaired osteoblast differentiation, diminished binding capacity of the osteoblast transcription factor Runx2, and a reduced number of osteocytes, highlighting the essential role of Lamin-A/C in maintaining bone health.
