Lactate dehydrogenase (LDH) is a crucial oxidoreductase enzyme primarily involved in the reversible conversion of pyruvate to lactate during anaerobic glycolysis. In mice, LDH activity is regulated by various factors, including hypoxia, which induces the expression of LDH-A and subsequently promotes lactate production. This metabolic shift, known as the "Warburg effect," is notably prominent in tumor cells, which often rely on increased glycolysis for energy production, even when sufficient oxygen is available. As mice age, the A/B isoform ratio of LDH in the brain shifts, leading to elevated lactate levels. This alteration is linked to mitochondrial dysfunction and neuronal damage, potentially accelerating cognitive decline.
