Target Information
Krueppel-like factor 1 (KLF1) is a transcriptional regulator crucial for erythrocyte development. It binds to the CACCC box in the beta-globin gene promoter to activate transcription and likely acts as a general switch for erythroid development. When sumoylated, KLF1 functions as a transcriptional repressor by facilitating interaction with CDH2/MI2beta and also inhibits megakaryocytic differentiation. In mice, KLF1 plays a vital role in erythropoiesis by regulating gene expression, promoting erythroid differentiation, and sustaining fetal liver hematopoiesis. It regulates the expression of erythroid genes by binding to the locus control region (LCR) and the promoter of the β-globin gene, in addition to interacting with histone acetyltransferases (HATs).
