Target Information
The KiSS-1 receptor (Kiss1r), also known as GPR54, is the receptor for metastin (kisspeptin-52 or kp-52), a C-terminally amidated peptide derived from the metastasis suppressor protein, KiSS1. Activation of this receptor suppresses cell proliferation and migration, key traits of tumor metastasis. In mice, Kiss1r plays a crucial role in the Kiss1 signaling pathway, primarily affecting reproductive and metabolic processes. Expressed in the hypothalamus, Kiss1r regulates the secretion of gonadotropin-releasing hormone (GnRH) by interacting with Kiss1 signals on GnRH neurons, thereby influencing reproductive function. Kiss1r signaling is essential for triggering puberty, secreting gonadal hormones, and ensuring normal reproductive function in adulthood. Mice lacking Kiss1r (Kiss1r KO) show reproductive dysfunction, including hypogonadotropic hypogonadism, mirroring the symptoms seen in humans with mutations in the Kiss1 or Kiss1r genes.
