Target Information
Interleukin-1 receptor type 2 (IL-1R2) is a non-signaling receptor for IL1A, IL1B, and IL1RN, and acts to diminish IL1B activities. It functions as a decoy receptor by competitively binding to IL1B, thereby preventing its interaction with IL1R1. Additionally, it modulates cellular response through a non-signaling association with IL1RAP after binding to IL1B. Both membrane-bound and secreted forms of IL1R2 preferentially bind IL1B and have lower affinity for IL1A and IL1RN. IL1R2 plays an anti-inflammatory and immunomodulatory role in mice by capturing IL-1 family cytokines to inhibit their signal transduction. Within the tumor microenvironment, IL1R2 is predominantly expressed in regulatory T cells (Tregs) and contributes to regulating tumor immune responses. In a colon cancer model, IL1R2-deficient mice demonstrated stronger anti-tumor immune responses and increased survival.
