Insulin receptor substrate 1 (IRS-1) is a signaling adapter protein involved in signal transduction from the insulin receptor (INSR) and insulin-like growth factor I receptor (IGF1R), playing a crucial role in development, growth, glucose homeostasis, and lipid metabolism. In mice, IRS-1 is pivotal in insulin signaling, regulating energy metabolism, growth and development, and disease resistance. Mice lacking IRS-1 exhibit abnormal glucagon secretion, leading to elevated blood sugar levels. Additionally, IRS-1 is essential for growth and development, as systemic IRS-1 knockout mice show growth retardation, weight loss, and abnormal bone development.
