Target Information

The insulin-degrading enzyme (IDE) is involved in breaking down insulin and various peptides such as APP peptides, IAPP peptides, natriuretic peptides, glucagon, bradykinin, kallidin, among others, thus playing a significant role in intercellular peptide signaling. When IDE binds to its substrates, it undergoes crucial conformational changes that allow it to bind and degrade larger substrates, like insulin. In mice, IDE's function extends beyond insulin degradation, involving the metabolic regulation of amyloid β and other peptides. This makes IDE a crucial target for research into metabolic and neurodegenerative diseases. IDE is particularly important in the breakdown of amyloid β, with studies indicating that IDE can reduce amyloid β accumulation in the brain, a key factor in Alzheimer's disease (AD) pathogenesis. For instance, in mouse models lacking the IDE gene, there is a substantial reduction in amyloid β degradation, leading to increased amyloid β levels in the brain, which aligns with AD's pathological characteristics.
