Target Information
High mobility group protein B1 (HMGB1) is a multifunctional redox-sensitive protein with diverse roles across various cellular compartments. In the nucleus, it is a key chromatin-associated non-histone protein that functions as a DNA chaperone, playing essential roles in replication, transcription, chromatin remodeling, V(D)J recombination, DNA repair, and maintaining genome stability. It is suggested to serve as a universal biosensor for nucleic acids. In mice, HMGB1 is primarily associated with inflammatory responses and immune regulation. Studies in mouse models have identified HMGB1 as a crucial late mediator of endotoxin lethality. Upon endotoxin stimulation, such as with lipopolysaccharide (LPS), HMGB1 release triggers macrophages to release large amounts of proinflammatory cytokines, including tumor necrosis factor (TNF-α) and interleukin (IL-1), leading to tissue damage and potentially fatal shock.
