Hepcidin is a liver-produced hormone that serves as the primary regulator of iron absorption and distribution throughout the body. It functions by promoting the endocytosis and degradation of SLC40A1, resulting in iron retention within iron-exporting cells and reduced iron flow into the plasma. In hepcidin knockout mice, increased expression of ferroportin, particularly in the liver and pancreas, causes iron overload, highlighting hepcidin's crucial role in maintaining iron homeostasis. Conversely, mice overexpressing hepcidin exhibit severe iron deficiency and anemia, further demonstrating its negative regulatory influence on iron balance.
