Growth arrest-specific protein 6 (Gas6) is a ligand for the tyrosine-protein kinase receptors AXL, TYRO3, and MER, which are involved in signaling pathways related to cell growth and survival, adhesion, and migration. GAS6/AXL signaling is crucial for various processes, such as preventing apoptosis in endothelial cells during acidification, optimizing cytokine signaling during human natural killer cell development, aiding hepatic regeneration, supporting the survival and migration of gonadotropin-releasing hormone neurons, activating platelets, and regulating thrombotic responses. In mice, Gas6 plays vital roles in promoting cell proliferation, regulating immune responses, participating in tissue repair, and influencing tumor development and metabolic processes. By binding to TAM family receptors like Axl, Mer, and Tyro3, Gas6 activates downstream signaling pathways, encouraging cell proliferation and migration. For instance, in vascular smooth muscle cells, Gas6 activates Ca2±dependent receptors to enhance cell proliferation.
