Target Information

In mice, Granulin (GRN) primarily functions in relation to prefrontal and temporal degenerative diseases, such as frontotemporal dementia (FTD). In a mouse model lacking PGRN (Grn^−/−), Granulin expression can significantly ameliorate neuropathological changes caused by PGRN deficiency, including inflammatory responses, lysosomal dysfunction, and synaptic loss. Granulin can normalize abnormal lysosomal protein levels, such as cathepsin Z and galectin-3, resulting from PGRN deficiency and improve lipid metabolism disorders linked to lysosomal function. By correcting lipid metabolism abnormalities, such as changes in BMP and GlcSph levels, Granulin helps prevent lipofuscin accumulation, a pathological characteristic of neurodegenerative diseases like FTD.
