Target Information

The glycine cleavage system facilitates the breakdown of glycine. The P protein (GLDC) attaches to the alpha-amino group of glycine using its pyridoxal phosphate cofactor, releasing CO2. The remaining methylamine group is transferred to the lipoamide cofactor of the H protein (GCSH). In mice, GLDC is involved not only in glycine metabolism but also in neural tube closure, folate metabolism, kidney development, and various physiological and pathological processes. Mouse models lacking GLDC, created through gene knockout techniques like GldcGT1/GT1, exhibit severe developmental issues, including craniofacial neural tube defects, hydrocephalus, growth retardation, and early death.
