Target Information

Glutamate-cysteine ligase catalyzes the ATP-dependent bonding of L-glutamate and L-cysteine, initiating and controlling the rate-limiting step in glutathione biosynthesis. In mice, Gclc is vital for synthesizing glutathione, regulating oxidative stress, and playing a crucial role in the development and pathology of various tissues. Mice with a liver-specific knockout of Gclc exhibit severe steatosis and liver failure. These mice begin to experience hepatocyte apoptosis and inflammatory responses by postnatal day 21 (PND21) and typically succumb to liver failure around PND30. Additionally, a complete loss of Gclc in the mouse central nervous system results in mitochondrial dysfunction and neurodegenerative diseases.
