Target Information
Glial cell line-derived neurotrophic factor (GDNF) is a neurotrophic factor that promotes the survival and morphological differentiation of dopaminergic neurons while enhancing their high-affinity dopamine uptake. It exerts its effects by binding to its coreceptor, GFRA1, which leads to autophosphorylation and activation of the RET receptor. GDNF plays a role in neural crest development. In mice, GDNF functions to protect dopaminergic neurons, contribute to Parkinson's disease treatment, regulate addictive behavior, and aid in nervous system development. It is expressed in the substantia nigra of adult mice, safeguarding dopaminergic neurons through RET and GFRα-1 receptor-mediated signaling pathways. Moreover, GDNF can decrease cocaine self-administration and alcohol intake in mice, possibly by regulating GABAergic neuron activity and enhancing immune reactivity influenced by ethanol.
