Target Information

Gelsolin is a calcium-regulated protein that modulates actin by attaching to the plus (or barbed) ends of actin monomers or filaments, thereby blocking monomer exchange (end-blocking or capping). It facilitates the assembly of monomers into filaments (nucleation) and can also sever already formed filaments. As a multifunctional actin regulatory protein, Gelsolin (GSN) is found in various cell types, including fibroblasts, neurons, erythrocytes, airway smooth muscle cells, and osteoclasts. Mice lacking Gelsolin (Gsn−/−) exhibit significantly reduced migration ability and increased Rac expression, but migration can be restored by introducing Gelsolin. Additionally, Gelsolin is critical for the formation of neuronal growth cones and the contraction of cilia. In Gelsolin-deficient mice, there is an increased number of cilia in neuronal growth cones and a reduced contraction rate, highlighting Gelsolin's essential role in neuronal morphogenesis.
