Target Information

Peptidyl-prolyl cis-trans isomerase FKBP5 is an immunophilin protein known for its PPIase and co-chaperone activities. It participates in unligated steroid receptor heterocomplexes by interacting with heat-shock protein 90 (HSP90). FKBP5 is involved in the intracellular trafficking of heterooligomeric forms of steroid hormone receptors, ensuring that the unliganded complex remains in the cytoplasm. FKBP5-deficient mice exhibit enhanced memory capabilities under acute stress conditions, but they also show significantly reduced cortisol levels, suggesting that FKBP5 indirectly influences memory and stress resilience by regulating glucocorticoid receptor (GR) activity. Additionally, the expression level of FKBP5 correlates with alcohol consumption and withdrawal behavior, underscoring its role in cognitive and behavioral regulation. FKBP5 expression is closely linked to the emotional state and anxiety-related behaviors in mice. Research has demonstrated that early life stress, such as separation stress, increases FKBP5 expression in the mouse brain, which may contribute to heightened anxiety and depression-like behaviors.
