Fibroblast growth factor 21 (FGF21) enhances glucose uptake in differentiated adipocytes by inducing the expression of glucose transporter SLC2A1/GLUT1, without affecting SLC2A4/GLUT4 expression. This activity likely requires the presence of KLB. FGF21 plays a vital role in regulating systemic glucose homeostasis and insulin sensitivity. In mice, FGF21 influences energy metabolism, lipid metabolism, insulin sensitivity, and liver health through various mechanisms, highlighting its potential as a target for treating obesity, diabetes, and related metabolic disorders. It is crucial in regulating energy metabolism, with studies showing that FGF21 can decrease body weight and body fat by increasing overall energy expenditure and physical activity. For example, in a diet-induced obesity mouse model, FGF21 treatment significantly reduced body weight and fat, while improving insulin sensitivity and liver steatosis. In wild-type female mice, ERα agonist treatment significantly elevated FGF21 expression and serum concentration, whereas in ERα knockout mice, FGF21 expression remained unchanged.
